Materials & Methods:
Live cell epifluorescence imaging was performed as described previously (25). Briefly NK-92 cells were cultured in glass-bottom 35-mm dishes (MatTek Ashland MA) coated with poly-L-lysine. Live cell epifluorescence imaging was performed using inverted microscope with a3100 oil objective (IX81 OBS real time; Olympus). Frame capture rates were between 500 and 2000 ms with total capture periods ranging from 2 to 20 min. Image control and postcapture image analysis were performed using CellR 2.5 software The Journal of Immunology 4853 (Olympus). Videos were analyzed cropped and constructed using ImageJ 1.37p (National Institutes of Health) and Volocity 3.7 (Improvision) and were exported as a QuickTime movie.
